Abstract：Fluorescent dyes are the cornerstones of avariety of modern biomedical technologies, including DNA sequenching,flowcytometry, fluorescence in-situ hybridization, and various forms of invitro or in vivo imaging modalities, etc. Development of novel fluorescentscaffolds and functional dyes to meet the ever-growing need from the biomedicalcommunity has been an ongoing research focus of synthetic dye community.
Ourgroup has a strong interest on xanthene dyes, a classic scaffold firstsynthesized by von Baeyer in the last 1870s. They have been tailored intolabels, probes, donors and more. However, development of novel molecular toolsbased on xanthenes may be limited by the available synthetic approaches. Overthe years, we have developed multiple new preparations of the xanthenescaffold. With these new preparations, we were able to access xanthenoids,which could not be prepared via alternative approaches.
Someof these xanthenoids exhibit strong absorption and emission in the nearinfrared or even infrared regaion, some are sterically protected to such anextent that it is not prone to nucleophilic attack by high concentration ofthiols or hydroxide; some have been rendered into a template forphoto-triggered drug-release; some are able to yield an intense fluorescence turn-onsignal in the presence of the analyte-of-interest; and some others haveexhibited potent anti-microbial activity against a range of multi-drug resistantpathogens.
